    Characterization of Mesenchymal Stem Cells in Mucolipidosis Type Ii (i-Cell Disease)

    Mucolipidosis type II (ML-II, I-cell disease) is a fatal inherited lysosomal storage disease caused by a deficiency of theenzyme N-acetylglucosamine-1-phosphotransferase. A characteristic skeletal phenotype is one of the many clinical manifestationsof ML-II. Since the mechanisms underlying these skeletal defects in ML-II are not completely understood, we hypothesized that adefect in osteogenic differentiation of ML-II bone marrow mesenchymal stem cells (BM-MSCs) might be responsible for this skeletalphenotype. Here, we assessed and characterized the cellular phenotype of BM-MSCs from a ML-II patient before (BBMT) and afterBM transplantation (ABMT), and we compared the results with BM-MSCs from a carrier and a healthy donor. Morphologically, wedid not observe differences in ML-II BBMT and ABMT or carrier MSCs in terms of size or granularity. Osteogenic differentiation wasnot markedly affected by disease or carrier status. Adipogenic differentiation was increased in BBMT ML-II MSCs, but chondrogenicdifferentiation was decreased in both BBMT and ABMT ML-II MSCs. Immunophenotypically no significant differences were observedbetween the samples. Interestingly, the proliferative capacity of BBMT and ABMT ML-II MSCs was increased in comparison to MSCsfrom age-matched healthy donors. These data suggest that MSCs are not likely to cause the skeletal phenotype observed in ML-II, butthey may contribute to the pathogenesis of ML-II as a result of lysosomal storage-induced pathology.

    Fitting a Recurrent Dynamical Neural Network To Neural Spiking Data: Tackling the Sigmoidal Gain Function Issues

    This is a continuation of a recent study (Doruk RO, Zhang K. Fitting of dynamic recurrent neural networkmodels to sensory stimulus-response data. J Biol Phys 2018; 44: 449-469), where a continuous time dynamical recurrentneural network is fitted to neural spiking data. In this research, we address the issues arising from the inclusion ofsigmoidal gain function parameters to the estimation algorithm. The neural spiking data will be obtained from the samemodel as that of Doruk and Zhang, but we propose a different model for identification. This will also be a continuoustime recurrent neural network, but with generic sigmoidal gains. The simulation framework and estimation algorithmsare kept similar to that of Doruk and Zhang so that we can have a solid base to compare the results. We evaluatethe estimation performance in two different ways. First, we compare the firing rate responses of the original and theestimated model. We find that responses of both models to the same stimuli are similar. Secondly, we evaluate variationsof the standard deviations of the estimates against a number of samples and stimulus parameters. They show a similarpattern to that of Doruk and Zhang. We thus conclude that our model serves as a reasonable alternative provided thatfiring rate is the response of interest (to any stimulus).

    Hydrogen Implantation Effects on the Electrical and Optical Properties of Inse Thin Films

    The effects of hydrogen ion implantation on the structural, electrical and optical properties of amorphous InSe thin films have been investigated. X-ray diffraction analysis revealed no change in the structure of the films. An implantation of 7.3 × $10^{18}$ ions/$cm^2$ decreased the electrical conductivity by three orders of magnitude at 300 K. Similarly, the conductivity activation energy, which was calculated in the temperature range of 300–420 K, decreased from 210 to 78 meV by H-ion implantation. The optical measurements showed that the direct allowed transitions energy band gap of amorphous InSe films has decreased from 1.50 to 0.97 eV by implantation. Furthermore, significant decreases in the dispersion and oscillator energy, static refractive index and static dielectric constants are also observed by hydrogen implantation.

    Neuron Modeling: Estimating the Parameters of a Neuron Model From Neural Spiking Data

    We present a modeling study aiming at the estimation of the parameters of a single neuron model from neural spiking data. The model receives a stimulus as input and provides the firing rate of the neuron as output. The neural spiking data will be obtained from point process simulation. The resultant data will be used in parameter estimation based on the inhomogeneous Poisson maximum likelihood method. The model will be stimulated by various forms of stimuli, which are modeled by a Fourier series (FS), exponential functions, and radial basis functions (RBFs). Tabulated results presenting cases with different sample sizes (# of repeated trials), stimulus component sizes (FS and RBF), amplitudes, and frequency ranges (FS) will be presented to validate the approach and provide a means of comparison. The results showed that regardless of the stimulus type, the most effective parameter on the estimation performance appears to be the sample size. In addition, the lowest variance of the estimates is obtained when a Fourier series stimulus is applied in the estimation.

    Ann-Assisted Forecasting of Adsorption Efficiency To Remove Heavy Metals

    In wastewater treatment, scientific and practical models utilizing numerical computational techniques suchas artificial neural networks (ANNs) can significantly help to improve the process as a whole through adsorption systems.In the modeling of the adsorption efficiency for heavy metals from wastewater, some kinetic models have been used such as pseudo first-order and second-order. The present work develops an ANN model to forecast the adsorption efficiency of heavy metals such as zinc, nickel, and copper by extracting experimental data from three case studies. To do this, we apply trial-and-error to find the most ideal ANN settings, the efficiency of which is determined by mean square error (MSE) and coefficient of determination (R2). According to the results, the model can forecast adsorption efficiency percent (AE%) with a tangent sigmoid transfer function (tansig) in the hidden layer with 10 neurons and a linear transferfunction (purelin) in the output layer. Furthermore, the Levenberg–Marquardt algorithm is seen to be most ideal for training the algorithm for the case studies, with the lowest MSE and high R2 . In addition, the experimental results and the results predicted by the model with the ANN were found to be highly compatible with each other.

    The Relationship Between Lung Function, Exercise Capacity, Oxidant and Antioxidant Response in Primary Ciliary Dyskinesia and Cystic Fibrosis

    Background. There is a need to identify the complex interplay between various physiological mechanisms in primary ciliary dyskinesia (PCD) and cystic fibrosis (CF). The study investigated the interaction between respiratory function, exercise capacity, muscle strength, and inflammatory and oxidant/antioxidant responses in patients with PCD and CF. Methods. The study included 30 PCD patients, 30 CF patients, and 29 age and sex-matched healthy subjects. Exercise capacity was assessed using the modified shuttle walk test (MSWT). Handgrip strength (HGS) was used to evaluate general muscle strength. Oxidative stress-inflammatory parameters were also assessed. Pulmonary function test was performed by spirometry. Regarding the forced expiratory volume in 1 second (FEV1) 1 ) z-score, patients with PCD and CF were subdivided into normal, mild, and severe/moderate groups. Results. Forced vital capacity (FVC) z-scores were lower in PCD and CF patients than controls. FEV1, 1 , FEV1/ 1 / FVC, peak expiratory flow (PEF), and forced mid expiratory flow (FEF 25-75% ) z-scores were lower in PCD than in the other groups. HGS was lower in both mild PCD and normal CF patients relative to the controls. MSWT distance was lower in severe/moderate PCD patients than controls. Catalase (CAT), glutathione S-transferase (GST), glutathione peroxidase (GPx), and malondialdehyde (MDA) levels did not differ significantly among the study groups, but superoxide dismutase (SOD) level in severe/moderate PCD, and glutathione (GSH) level in normal CF were higher than in controls. Interleukin-6 (IL-6) level was higher in patients with normal PCD and CF compared to the controls. IL-1 beta level was higher in PCD compared to controls. Additionally, correlations among these parameters were also determined in some patient groups. Conclusion. Homeostasis related to respiratory function, aerobic performance, muscle strength, inflammatory response, and oxidant/antioxidant balance were affected in PCD and CF. Evaluating these mechanisms together may contribute to elucidating the pathophysiology of these rare diseases.

    The Effects of Eryngium Campestre Extracts on Glutathione-S Glutathione Peroxidase and Catalase Enzyme Activities;

    Eryngium campestre L. (Apiaceae) are known in Turkish folk medicine as ‘Bogadikeni’ and wildly distributed in all parts of Turkey. Infusion of different parts of this species are well known as diuretic, appetizer and stimulant. In the present study, methanol extracts from flowers and leaves of E. campestre were evaluated for their potential medicinal value in terms of biological targets participate in antioxidant defense such as glutathione-S-transferase (GST), glutathione peroxidase (GPx) and catalase (CAT). Results obtained show that the methanol extract of flowers were found to have higher amount of phenolic and flavonoid contents than the leaves. The evaluation of extracts against biological target revealed that, the maximum GST enzymes inhibition activities was observed with methanol extract from flowers of E. campestre with IC50 value of 363 ng/mL. In this study, none of the extracts evaluated were shown any reasonable GPx and CAT inhibition. © 2014, Turkish Pharmacists Association. All rights reserved.

    An Approach for Performance Prediction of Saturated Brushed Permanent Magnet\rdirect Current (dc) Motor From Physical Dimensions

    An analytical approach for performance prediction of saturated brushed permanent magnet direct current\r(DC) motors is proposed in this paper. In case of a heavy saturation in the stator back core of electrical machines, some\rflux completes its path through the surrounding air, and the conventional equivalent circuit cannot be used anymore.\rThis issue has not been addressed in the literature. The importance of considering the effect of the flux penetrating\rthe surrounding air is shown in this paper using finite element simulations and experimental results, and an analytical\rapproach is proposed to consider this effect on magnet operating point determination and performance prediction of\rsaturated brushed permanent magnet DC motors. An analytical method is also presented to determine the boundary\rradius of the surrounding air for obtaining accurate results in finite element (FE) solutions and analytical calculations.\rAn analytical approach based on Carter’s coefficient is also proposed to calculate the effective length of the magnet when\rthe length of the magnet and rotor length are not the same. The accuracy of the proposed analytical model is illustrated\rusing finite element simulations and experimental results. With this accuracy, this analytical model is very suitable to\rbe used for reliable and quick mathematical design optimization.

    Bağıntı ve Fonksiyonlar Konusunda Yapılan Yaygın Hataların Belirlenmesi ve Giderilmesi Üzerine Boylamsal Bir Çalışma

    Bu çalışma boylamsal bir çalışmadır. Araştırmanın birinci basamağı, 2004–2005 öğretim yı-lında Hacettepe Üniversitesi Sınıf Öğretmenliği Bölümü’nde birinci sınıfta okumakta olan 190 öğrenci üzerinde uygulanmıştır. Bu aşamada, öğrencilerin bağıntı ve fonksiyonlar konusunda yaptıkları yaygın hatalar belirlenmiş, sınıflandırılmış ve bu hataların temelinde yatan nedenler araştırılmıştır. Araştırmanın ikinci basamağı, 2005–2006 öğretim yılında aynı üniversitenin aynı bölümünde ve yine birinci sınıfta okumakta olan 97 öğrenci ile yürütülmüştür. Bu aşamada, “Sınıf öğretmenliği öğrencilerinin fonksiyonlar konusunda yaptıkları yaygın hatalar giderilebilir mi?” sorusuna yanıt aranmaktadır. Çalışmanın sonuçları, birinci aşamada saptanan yaygın hatalar dikkate alınarak hazırlanmış olan ders içeriği, dersin işleniş planı ve kullanılan metot ile hataların büyük ölçüde giderildiğini göstermiştir.

    From Partners To Rivals: Normative Power Europe Meets Normative Power China

    The European Union (EU) has been promoting its norms, values, and rules for decades. However, in the current international environment, the EU’s normative power is not being received well outside the EU, particularly in reference to the growing power of illiberal states. Within that context, this study explores EU-China relations across time and unpacks the position of normative power Europe towards China and the Chinese response. The study foregrounds the fruitless attempts of the EU to project its transformative power onto China and the growing resistance by China against this, which it expresses by presenting itself to the West as an alternative power with an alternative understanding of international politics.
