In this study, the distribution of the dominant bacteria present in the microbiota of herby cheese samples (n = 13) were examined by the next generation sequencing (NGS) technique. DNA was extracted both directly from cheese samples and after pre-enrichment. The metagenomic analysis of the NGS results revealed that Firmicutes were dominant both in DNA directly extracted from herby cheese (KOP), and pre-enriched samples (OP), at the phylum level. At the genus level, Lactobacillus, Lactococcus, and Streptococcus were dominant in the KOP samples, whereas in the OP samples, Enterococcus, Streptococcus, and Bacillus were determined as the dominant bacterial genera. Although Lactococcus raffinolactis and Streptococcus salivarius were dominant in the KOP samples, Enterococcus faecalis and S. salivarius were dominant in the OP samples. The Shannon species diversity index and principal coordinates analysis (PCoA) were used to determine the distribution in KOP and OP samples at the genus level. The PCoA of KOP-10, KOP-11, KOP-2, and KOP-7, KOP-3, and KOP-6 samples showed the wide distribution, whereas KOP-5, KOP-8, KOP-9, and KOP-14 herby cheese samples were closely related. The OP samples, especially OP-7 and OP-14, showed wide distribution in comparison to other OP samples. Finally, the dominant bacterial communities were identified by DNAbased metagenomic analysis, and this is the first report to elucidate the microbiota of herby cheese produced in Turkey using the NGS technique.Article Citation Count: 12-Ag and Bone Marrow-Targeted Pcl Nanoparticles as Nanoplatforms for Hematopoietic Cell Line Mobilization(Bmc, 2024) Kose, Sevil; Varan, Cem; Onen, Selin; Nemutlu, Emirhan; Bilensoy, Erem; Korkusuz, PetekBackgroundThe use of mobilizing agents for hematopoietic stem cell (HSC) transplantation is insufficient for an increasing number of patients. We previously reported lipid made endocannabinoid (eCB) ligands act on the human bone marrow (hBM) HSC migration in vitro, lacking long term stability to be therapeutic candidate. In this study, we hypothesized if a novel 2-AG-loaded polycaprolactone (PCL)-based nanoparticle delivery system that actively targets BM via phosphatidylserine (Ps) can be generated and validated.MethodsPCL nanoparticles were prepared by using the emulsion evaporation method and characterized by Zetasizer and scanning electron microscopy (SEM). The encapsulation efficiency and release profile of 2-AG were determined by liquid chromatography-tandem mass spectrometry (LC-MS/MS). The presence of cannabinoid receptors (CBRs) in HSCs and monocytes was detected by flow cytometry. Cell morphology and viability were assessed using transmission electron microscopy (TEM), SEM, and the WST-1 viability assay. The migration efficacy of the 2-AG and 2-AG-loaded nanoparticle delivery system on HSCs and HPSCs (TF-1a and TF-1) and monocytes (THP-1) was evaluated using a transwell migration assay.ResultsThe 140-225 nm PCL nanoparticles exhibited an increasing polydispersity index (PDI) after the addition of Ps and 2-AG, with a surface charge ranging from - 25 to -50 mV. The nanoparticles released up to 36% of 2-AG within the first 8 h. The 2-AG-Ps-PCL did not affect cellular viability compared to control on days 5 and 10. The HSCs and monocytes expressed CB1R and CB2R and revealed increased migration to media containing 1 mu M 2-AG-Ps-PCL compared to control. The migration rate of the HSCs toward monocytes incubated with 1 mu M 2-AG-Ps-PCL was higher than that of the monocytes of control. The 2-AG-Ps-PCL formulation provided a real time mobilization efficacy at 1 mu M dose and 8 h time window via a specific CBR agonism.ConclusionThe newly generated and validated 2-AG-loaded PCL nanoparticle delivery system can serve as a stable, long lasting, targeted mobilization agent for HSCs and as a candidate therapeutic to be included in HSC transplantation (HSCT) protocols following scale-up in vivo preclinical and subsequent clinical trials.Article Citation Count: 53-D Dynamic Uav Base Station Location Problem(informs, 2021) Cicek, Cihan Tugrul; Çiçek, Cihan Tuğrul; Shen, Zuo-Jun Max; Gultekin, Hakan; Tavli, Bulent; Industrial EngineeringWe address a dynamic covering location problem of an unmanned aerial vehicle base station (UAV-BS), in which the location sequence of a single UAV-BS in a wireless communication network is determined to satisfy data demand arising from ground users. This problem is especially relevant in the context of smart grid and disaster relief. The vertical movement ability of the UAV-BS and nonconvex covering functions in wireless communication restrict utilizing classical planar covering location approaches. Therefore, we develop new formulations to this emerging problem for a finite time horizon to maximize the total coverage. In particular, we develop a mixed-integer nonlinear programming formulation that is nonconvex in nature and propose a Lagrangean decomposition algorithm (LDA) to solve this formulation. Because of the high complexity of the problem, the LDA is still unable to find good local solutions to large-scale problems. Therefore, we develop a continuum approximation (CA) model and show that CA would be a promising approach in terms of both computational time and solution accuracy. Our numerical study also shows that the CA model can be a remedy to build efficient initial solutions for exact solution algorithms. Summary of Contribution: This paper addresses a facet of mixed integer nonlinear programming formulations. Dynamic facility location problems (DFLPs) arise in a wide range of applications. However, classical DFLPs typically focus on the two-dimensional spaces. Emerging technologies in wireless communication and some other promising application areas, such as smart grids, have brought new location problems that cannot be solved with classical approaches. For practical reasons, many research attempts to solve this new problem, especially by researchers whose primary research area is not OR, have seemed far from analyzing the characteristics of the formulations. Rather, solution-oriented greedy heuristics have been proposed. The ultimate goal of bioprinting is to produce stable, anatomically-shaped, human-scale functional organs or tissue substitutes that can be implanted. Although various bioprinting techniques have emerged to develop customized tissue-engineering substitutes over the past decade, several challenges remain in fabricating volumetric tissue constructs with complex shapes and sizes and translating the printed products into clinical practice. Thus, it is crucial to develop a successful strategy for translating research outputs into clinical practice to address the current organ and tissue crises and improve patients' quality of life. This review article discusses the challenges of the existing bioprinting processes in preparing clinically relevant tissue substitutes. It further reviews various strategies and technical feasibility to overcome the challenges that limit the fabrication of volumetric biological constructs and their translational implications. Additionally, the article highlights exciting technological advances in the 3D bioprinting of anatomically shaped tissue substitutes and suggests future research and development directions. Cannabinoid ligands exert antiproliferative effect and induce apoptosis on numerous epithelial cancers. We confirmed that CB1 receptor (CB1R) is expressed in NSCLC cells in this study. Arachidonoylcyclopropylamide (ACPA) as a synthetic, CB1R-specific ligand decreased proliferation rate in NSCLC cells by WST-1 analysis and real-time proliferation assay (RTCA). The half-maximal inhibitory concentration (IC50) dose of ACPA was calculated as 1.39x10(-12)M. CB1 antagonist AM281 inhibited the antiproliferative effect of ACPA. Flow cytometry and ultrastructural analyzes revealed significant early and late apoptosis with diminished cell viability. Nano-immunoassay and metabolomics data on activation status of CB1R-mediated pro-apoptotic pathways found that ACPA inhibited Akt/PI3K pathway, glycolysis, TCA cycle, amino acid biosynthesis, and urea cycle and activated JNK pathway. ACPA lost its chemical stability after 24hours tested by liquid chromatography-mass spectrometry (LC-MS/MS) assay. A novel ACPA-PCL nanoparticle system was developed by nanoprecipitation method and characterized. Sustained release of ACPA-PCL nanoparticles also reduced proliferation of NSCLC cells. This suggests that the free carrier activation mechanisms form a continuum in a large temperature scale (300 K - 858 K) and the metastable amorphous phase can be treated as a supercooled liquid. The effective activation energy calculated using the resistivity versus temperature data follow a parabolic behavior, with a room temperature value of 333 meV, peaking to similar to 377 meV at similar to 465 K and reaching zero at similar to 930 K, using a reference activation energy of 111 meV (3k(B)T/2) at melt. Amorphous GST is expected to behave as a p-type semiconductor at T-melt similar to 858 K and transitions from the semiconducting-liquid phase to the metallic-liquid phase at similar to 930 K at equilibrium. The simultaneous Seebeck (S) and resistivity versus temperature measurements of amorphous-fcc mixed-phase GST thin-films show linear S-T trends that meet S = 0 at 0 K, consistent with degenerate semiconductors, and the dS/dT and room temperature activation energy show a linear correlation. The single-crystal fcc is calculated to have dS/dT = 0.153 mu V/K-2 for an activation energy of zero and a Fermi level 0.16 eV below the valance band edge. The English version of inventory consists of 4 sub-factors that measure cravings for high-fat foods, carbohydrates/starches, sweets, and fast food fats, and creates a total score. The sample of the study consists of 621 individuals between the ages of 19-50 who voluntarily agree to participate in an online survey. This study revealed that FCI-TR is a valid instrument of specific food cravings in the Turkish adult population. Turkish version of the FCI also consist of 4 sub-factors. Women experienced more food craving for sweets than men. While the most craved food by women was chocolate, men scored significantly higher on bread than women. In addition, a relationship was found between food craving and body weight. Introduction The Food Craving Inventory is a 28-item self-report measure of specific food cravings. The inventory consists of 4 factors: high fats, sweets, carbohydrates/starches and fast-food fats. Purpose This study was carried out to evaluate the Turkish validity and reliability of the Food Craving Inventory, and to determine the psychometric properties and factor structure of the Turkish version. Methods The sample of the study consists of 621 individuals between the ages of 19-50 who voluntarily agree to participate in online survey. Validity and reliability analyses were performed for the Turkish version of Food Craving Inventory (FCI-TR). Confirmatory factor analysis was performed to evaluate the factor structure of the Turkish version of FCI. Results Confirmatory factor analysis yielded a four-factor structure as "sweets," "high-fats," "carbohydrates/starches" and "fast food fats". The Cronbach-alpha coefficient for the total score was 0.84; subfactors were calculated as 0.74 for "sweets", 0.64 for "high-fat foods", 0.65 for "carbohydrates/starches", and 0.66 for "fast-food fats". The scores of the FCI-TR factors and its total score significantly correlated with the sub-factors of Three Factor Eating Questionnaire (TFEQ). A significant correlation was found between body mass index (BMI) and high fats and fast-food fats factor score. Also total and factor scores of the FCI-TR were different between BMI groups. Conclusions This study demonstrates that the Turkish version of the FCI is a valid and reliable tool to measure food cravings in the Turkish population. For evaluating the integrals in each subdivision, Gauss quadrature is applied. The method is computationally simple, memory efficient and can be applied for both triangular and quadrilateral elements, including the elements with nonplanar and/or curved surfaces. To assess the method, several examples are discussed. Although stimuli that are optimized for model parameter estimation should, in theory, have advantages over nonadaptive random stimuli, in practice it remains unclear in what way and to what extent an optimal design of time-varying stimuli may actually improve parameter estimation for this common type of recurrent network models. Here we specified the time course of each stimulus by a Fourier series whose amplitudes and phases were determined by maximizing a utility function based on the Fisher information matrix. To facilitate the optimization process, we have derived differential equations that govern the time evolution of the gradients of the utility function with respect to the stimulus parameters. The network parameters were estimated by maximum likelihood from the spike train data generated by an inhomogeneous Poisson process from the continuous network state. The adaptive design process was repeated in a closed loop, alternating between optimal stimulus design and parameter estimation from the updated stimulus-response data. Our results confirmed that, compared with random stimuli, optimally designed stimuli elicited responses with significantly better likelihood values for parameter estimation. Furthermore, all individual parameters, including the time constants and the connection weights, were recovered more accurately by the optimal design method. We also examined how the errors of different parameter estimates were correlated, and proposed heuristic formulas to account for the correlation patterns by an approximate parameter-confounding theory. Our results suggest that although adaptive optimal stimulus design incurs considerable computational cost even for the simplest excitatory-inhibitory recurrent network model, it may potentially help save time in experiments by reducing the number of stimuli needed for network parameter estimation.Article Citation Count: 31Adoption of Mobile Applications for Teaching-Learning Process in Rural Girls' Schools in India: an Empirical Study(Springer, 2020) Chatterjee, Sheshadri; Mısra, Sanjay; Majumdar, Dipasree; Misra, Sanjay; Damasevicius, Robertas; Computer EngineeringThe purpose of this study is to identify the factors that can impact the adoption of mobile apps for teaching-learning process focusing on the girls' school in rural India. The hypotheses were proposed and a conceptual model has been developed. There is a survey work conducted to collect the data from different respondents using a convenience sampling method. The model has been validated statistically through PLS-SEM analysis covering feedbacks of 271 effective respondents. The study highlights the impact of different antecedents of the behavioural intention of the students of using mobile applications for teaching-learning process. The results also show that among other issues, price value has insignificant influence on the intention of the girl students of the rural India. During survey feedbacks have been obtained from the 271 respondents, which is meagre compared to vastness of the population and school of rural India. Only few predictors have been considered leaving possibilities of inclusion of other boundary conditions to enhance the explanative power more than that has been achieved in the proposed model with the explanative power of 81%. The model has provided laudable inputs to the educational policy makers and technology enablers and administrators to understand the impact of the mobile applications on the rural girls' school of India and facilitate the development of m-learning. AE is defined as the nonformal education for individuals aged older than 25 years. The outcome of AE is measured by changing jobs for employed and finding a job for the unemployed. Concentrating on employed people, we analyze both the determinants and the outcome of participation in AE for the purpose of changing jobs; and second, concentrating on unemployed people, we analyze both the determinants and the outcome of participation in AE for the purpose of getting employed. We find that once young males who are already working participate in AE for changing work, independent of their education or how AE is financed, they can be successful in doing so. Leveraging the software-defined architecture, this study encompasses the intricacies of frequency modulated continuous waveform (FMCW) design for three distinct stages of UAS flight: cruise, landing approach, and touchdown within a signal processing framework. Angle of arrival (AoA) estimation, traditionally employed in terrain mapping applications, is largely unexplored for UAS radar altimeters (RAs). Time-division multiplexing multiple input-multiple output (TDM-MIMO) is an efficient method for enhancing angular resolution without compromising the size, weight, and power (SWaP) characteristics. Accordingly, this work argues the potential of AoA estimation using TDM-MIMO to augment situational awareness in challenging landing scenarios. To this end, two corner cases comprising landing a small-sized drone on a platform in the middle of a water body are included. Likewise, for the touchdown stage, an improvised rendition of zoom fast Fourier transform (ZFFT) is investigated to achieve millimeter (mm)-level range accuracy. Aptly, it is proposed that a mm-level accurate RA may be exploited as a software redundancy for the critical weight-on-wheels (WoW) system in fixed-wing commercial UASs. Each stage is simulated as a radar scenario using the specifications of automotive radar operating in the 77-81 GHz band to optimize waveform design, setting the stage for field verification. This article addresses challenges arising from radial velocity due to UAS descent rates and terrain variation through theoretical and mathematical approaches for characterization and mandatory compensation. While constant false alarm rate (CFAR) algorithms have been reported for ground detection, a comparison of their variants within the scope UAS altimetry is limited. Our results show Nigeria's current agricultural state, threats to food security, and modern digital agriculture technologies. We adapted our SLR findings to develop an implementation framework for agriculture 4.0 in solving Nigeria's food insecurity challenge in the post-Covid-19 era. Our proposed framework integrates precision agriculture in Nigeria's food production and the necessary enabling digital technologies in the agri-food supply chain. We analyzed the critical implementation considerations during each agri-food supply chain stage of farming inputs, farming scale, farming approach, farming operation, food processing, food preservation/storage, distribution/logistics, and the final consumers. This study will help researchers, investors, and the government address food security in Nigeria. In the period after 2nd World War, big cities in Turkey, like many other countries, entered in the process of rapid urbanization under the influence of mechanization in agriculture and migration from rural to urban areas. Increase in the population of cities and scarcity of shelter brought with the emergence of squatter areas. Batikent settlement was developed as a flagship solution to this housing problem and established a precedent with its model of implementation for future urban cohabitations. Batikent, as a new settlement area, was initiated by Municipality of Ankara and developed a never-before-seen operational model with three main collaborators: central government, municipality and association of housing cooperatives (Kent-Koop). Separation of roles - such as passing fiscal and housing laws, providing financial support, expropriation of land, constructing technical infrastructure, establishing municipal and commercial services, preparing master plan and designing site and house plans - through these collaborators provided this new mass development project with effective, practical and economical solutions. This study will focus on how Batikent was developed as a platform for innovation with the operational model based on dialogue between different parties. Collection of plans from master plans to layouts of apartments, meeting notes and feasibility studies documented and published by Kent-Koop will be used in this study to achieve a better understanding of the planning process of this exemplary quarter.Article Citation Count: 1The Amplification of the New Keynesian Models and Robust Optimal Monetary Policy(Amer inst Mathematical Sciences-aims, 2020) Ozcan, Gulserim; Akbal, Gülserim Özcan; EconomicsThis paper analyzes whether and how model uncertainty affects the amplification mechanism of the New Keynesian models in a simple min-max framework where the central bank plays a zero-sum game versus a hypothetical, evil agent. A first finding on a benchmark model with staggered price setting is that a robust optimal commitment policy necessitates more aggressive policy under a demand shock. Further, bringing additional persistence into the model deteriorates the effectiveness of monetary policy. Hence, allowing for either habit formation or partial indexation of prices to lagged inflation rate requires a stronger response for the policy to a demand shock. Together with the specification doubts, in order to reassure the private sector and signal that it will stabilize the fluctuations in the output gap, the policymaker reacts more aggressively as persistence rises. Although inflation persistence does not change the impact of model uncertainty, habit formation in consumption eliminates even reverses the impact of uncertainty on the policy reaction to a supply shock. In all cases, policymaker attributes less importance to nominal interest rate inertia with concerns about model uncertainty.
