    Characterization of Mesenchymal Stem Cells in Mucolipidosis Type Ii (i-Cell Disease)

    Mucolipidosis type II (ML-II, I-cell disease) is a fatal inherited lysosomal storage disease caused by a deficiency of theenzyme N-acetylglucosamine-1-phosphotransferase. A characteristic skeletal phenotype is one of the many clinical manifestationsof ML-II. Since the mechanisms underlying these skeletal defects in ML-II are not completely understood, we hypothesized that adefect in osteogenic differentiation of ML-II bone marrow mesenchymal stem cells (BM-MSCs) might be responsible for this skeletalphenotype. Here, we assessed and characterized the cellular phenotype of BM-MSCs from a ML-II patient before (BBMT) and afterBM transplantation (ABMT), and we compared the results with BM-MSCs from a carrier and a healthy donor. Morphologically, wedid not observe differences in ML-II BBMT and ABMT or carrier MSCs in terms of size or granularity. Osteogenic differentiation wasnot markedly affected by disease or carrier status. Adipogenic differentiation was increased in BBMT ML-II MSCs, but chondrogenicdifferentiation was decreased in both BBMT and ABMT ML-II MSCs. Immunophenotypically no significant differences were observedbetween the samples. Interestingly, the proliferative capacity of BBMT and ABMT ML-II MSCs was increased in comparison to MSCsfrom age-matched healthy donors. These data suggest that MSCs are not likely to cause the skeletal phenotype observed in ML-II, butthey may contribute to the pathogenesis of ML-II as a result of lysosomal storage-induced pathology.

    Identification of Bacterial Diversity of Bee Collected Pollen and Bee Bread Microbiota by Metagenomic Analysis

    This study investigated the bacterial diversities of bee-collected pollen and bee bread of Apis mellifera in Turkey. The bacterial community structure of 14 bee pollen from Bingol, Konya, and Hakkari and 11 bee bread samples from Bingol were studied using 16 S rRNA amplicon sequencing and metagenomic analysis. The dominant bacterial phylum in pollen and bee bread samples was Firmicutes, followed by Proteobacteria. In pollen and bee bread samples, Bacillaceae, Clostridiaceae, Enterococcaceae, and Enterobacteriaceae were identified as dominant bacterial families. At the genus level, Bacillus, Clostridium sensu stricto, and Enterococcus were dominant bacteria in both pollen and bee bread samples. The most abundant species was Clostridium perfringens in both pollen and bee bread samples. Escherichia vulneris, Enterococcus faecalis, Bacillus cereus, Enterococcus casseliflavus, and Cronobacter malonaticus were identified with high reads in pollen samples. In bee bread samples, E. faecalis, Clostridium bifermentans, and Pantoea calida were abundant bacterial species. Alpha diversity showed that pol-3 sample had the highest diversity. Beta-diversity plots separated the pollen samples into four main groups and bee bread samples into three main groups. Our results indicated that the culture-independent metagenomic analysis will be a valuable tool for determining the microbial diversity of bee products produced in Bingol-Turkey one of the important centers of apiculture.

    On the Yielding of Two-Layer Composite Spherical Pressure Vessels

    The yielding of two-layer composite spherical pressure vessels under either internal or external pressure is investigated analytically in the framework of small deformations and von Mises yield criterion. It is shown for both pressure cases that depending on the material properties and sphere dimensions, different modes of plasticization may take place. Unlike the deformation behavior of a single layer spherical pressure vessel, yielding may commence at the inner layer or at the outer layer or simultaneously at both layers of the assembly.

    Current Induction Therapy Strategies and Anti-T Lymphocyte Globulin Usage in Kidney Transplantation: Consensus-Based Recommendations by a Turkish Expert Panel

    This advisory committee convened to review national and global kidney transplantation dynamics and provide recom- mendations on the use of anti-T lymphocyte globulin (ATLG) for prevention and treatment of rejection after allogeneic kidney transplantation. A critical evaluation of 6 relevant articles released up to October 2022 was performed to reveal their importance in clinical practice. Additionally, 27 key questions on the indication, dosage of ATLG, and risk stratification were used for the Delphi technique with 8 members of the Turkish Society of Nephrology including 5 kidney transplanta- tion (KTx) subcommittee members and a surgeon experienced in solid organ transplantation. The committee declared that Türkiye had great potential in KTx; however, increase in transplantation would be possible in the case of raise in the deceased donor transplantation. As a consensus, ATLG was strongly recommended for induction and rejection treatment. Also, committee members recommended the safe dosage range in steroid resistant acute rejection as 2.5-3 mg/kg daily for 5-7 days, and the median of preferred dosage in induction sounded as 2-2.5 mg/kg daily for 3 days in intermediate risk state. Additionally, post-transplant infection and malignancy cases due to immunosuppression were much rarely encoun- tered than they were in the past.

    Surgical Treatment of a Patient With Recurrent Bleb Leak and Glaucoma: Bleb Excision Combined With Gonioscopy-Assisted Transluminal Trabeculotomy

    Here we present a case of intermittent bleb leakage with increased intraocular pressure (IOP) during recovery periods that was treated with gonioscopy-assisted transluminal trabeculotomy (GATT) combined with avascular bleb excision. A 60-year-old woman exhibiting simultaneous leaking bleb and glaucoma underwent GATT and bleb revision. At her final visit, the bleb leakage had resolved and IOP was under control without any further antiglaucoma medication. GATT may be useful for glaucoma patients exhibiting intermittent bleb leakage after failed trabeculectomy.

    Borulu sulama şebekeleri ülkelerin tarımsal ürün yetiştirmesinde ve su tasarrufunda enönemli sistemlerdir. Günümüzde tarımsal amaçlı sulama, su tüketiminin en yüksek olduğusektördür. Ülkemizde tarım sektörünün kullandığı su miktarı su potansiyelimizin %73oranındadır. Ülkemizde ve genelde Dünyada nüfus artışı nedeniyle tarımsal üretimde sutalebinin artması ve su kaynaklarımızın sınırlı olması nedeniyle sürdürülebilir su kaynaklarıyönetimi zorunlu olmaktadır. Son yıllarda ülkemizde yapılan sulama sistemleri yatırımlarınınbüyük bir çoğunluğunu, su kaybını ve tarımsal su kullanımını azaltan modern borulu sulamaşebekeleri yatırımları oluşturmaktadır. Bu çalışma borulu sulama şebekelerinintasarımlarının maliyet ve istenen işletme basınçları açısından optimum koşullara ulaşmasınaodaklanmıştır. Optimizasyon problemi çözümünde heuristik tabanlı dinamik programlamaile birlikte doğrusal programlama yöntemi olan “Simplex” metodu kullanılmış olup, buoptimizasyon yazılımı DSİ borulu sulama şebekeleri için uygulanmıştır.

    Fitting a Recurrent Dynamical Neural Network To Neural Spiking Data: Tackling the Sigmoidal Gain Function Issues

    This is a continuation of a recent study (Doruk RO, Zhang K. Fitting of dynamic recurrent neural networkmodels to sensory stimulus-response data. J Biol Phys 2018; 44: 449-469), where a continuous time dynamical recurrentneural network is fitted to neural spiking data. In this research, we address the issues arising from the inclusion ofsigmoidal gain function parameters to the estimation algorithm. The neural spiking data will be obtained from the samemodel as that of Doruk and Zhang, but we propose a different model for identification. This will also be a continuoustime recurrent neural network, but with generic sigmoidal gains. The simulation framework and estimation algorithmsare kept similar to that of Doruk and Zhang so that we can have a solid base to compare the results. We evaluatethe estimation performance in two different ways. First, we compare the firing rate responses of the original and theestimated model. We find that responses of both models to the same stimuli are similar. Secondly, we evaluate variationsof the standard deviations of the estimates against a number of samples and stimulus parameters. They show a similarpattern to that of Doruk and Zhang. We thus conclude that our model serves as a reasonable alternative provided thatfiring rate is the response of interest (to any stimulus).

    Covid-19 Infection in Peritoneal Dialysis Patients: \ra Comparative Outcome Study With Patients on \rhemodialysis and Patients Without Kidney Disease

    Objectives: There is limited data about coronavirus disease-19 (COVID-19) characteristics and results in peritoneal dialysis \r(PD) patients. This study aimed to investigate the characteristics and outcomes among PD patients and compare them with \rmatched hemodialysis (HD) patients and a control group without kidney disease.\rMethods: We included 18 PD patients and consecutive age- and gender-matched 18 HD and 18 patients without kidney \rdisease (control group) registered into the Turkish Society of Nephrology database including 1301 COVID-19 patients. We \rcompared demographic, clinical, radiological, laboratory data, and outcomes namely intensive care unit (ICU) admission, \rmechanical ventilation, mortality, and composite outcome (death and/or ICU admission).\rResults: ICU admission, mechanical ventilation, and mortality rates in PD patients (27.8%, 22.2%, and 22.2%, respectively) \rand the HD group (16.7%, 11.1%, and 16.7%, respectively) were higher than the control group (11.1%, 11.1%, and 5.6%, \rrespectively), but intergroup comparison did not reveal difference. A total of 11 (20.3%) patients had composite outcome \r(6 PD patients, 3 HD patients, and 2 patients in the control group). In Cox regression analysis, higher age and higher CRP level were related to increased risk of composite outcome. Adjusted rate of composite outcome in PD group was significantly higher than \rthe control group (P = .050). This rate was similar in HD and control groups (P = .30).\rConclusions: Combined in-hospital mortality and/or ICU admission of PD patients with COVID-19 was significantly higher than the control \rpatients. There is a need for careful surveillance of PD patients for infection signs and prompt treatment of COVID-19.

    Nöromüsküler Hastalarda Gövde Kontrolünün Değerlendirilmesi: Gövde Bozukluk Ölçeği’nin Geçerlik ve Güvenirliği

    Amaç: Çalışmamızın amacı Gövde Bozukluk Ölçeği’nin (GBÖ) erişkin nöromüsküler hastalıklarda geçerlik ve güvenirliğini araştırmaktır. Gereç ve Yöntem: Çalışmaya 66 nöromüsküler hastalık tanısı olan birey alındı. Çalışmaya dahil edilen olgulara GBÖ, Manuel Kas Testi (gövde, üst ve alt ekstremite), Motor Fonksiyon Değerlendirme Ölçeği (MFM), Fonksiyonel Bağımsızlık Ölçeği (FIM), Rivermead Mobilite İndeksi (RMI) yapıldı. Bulgular: GBÖ’nün test tekrar test, ICC güvenirlikleri mükemmel bulundu (GBÖ: 0,96 ve 0,98). Cronbach alfa katsayısı 0,82 bulundu. Yapı geçerliği için GBÖ ile MFM (r=0,29), MFM gövde (r=0,31), toplam gövde kas kuvveti (r=0,34), toplam alt ekstremite kas kuvveti (r=0,34, p<0,05), toplam kas kuvveti (gövde, alt ve üst ekstremite) (r=0,52, p<0,05) arasında anlamlı ilişki bulundu. GBÖ uyum geçerliği için FIM (r=0,32, p<0,05) ve RMI (r=0,39, p<0,05) ile ilişkili bulundu. Sonuç: Çalışmanın sonucunda GBÖ’nün erişkin kas hastalıklarında yapı ve uyum geçerliliğinin olduğu ve mükemmel güvenirliğe sahip olduğu bulundu. GBÖ rehabilitasyon kliniklerinde uygulaması kolay ve kısa zaman alan bir ölçektir

    Evaluation of Students' Opinions Regarding Distance Learning Practices in Turkish Universities During the Covid-19 Pandemic

    The purpose of this study is to identify hether students at Turkish universities are having difficulties in the distance learning process and to reveal their opinions regarding the system. Using a cross-cultural survey design, data were collected from 5052 students. Based on the collected data, 58.5% of the students have asynchronous distance learning classes in their universities. Access to distance learning classes is one of the problems, and it is attributed to computers, internet, and connectivity issues as well as with starting the lessons too early or late. A majority of participants (73.7%) found class materials uploaded by lecturers to distance learning systems to be sufficient or partially sufficient, and 36.7% stated that they had problems accessing resources regarding their homework. Students' satisfaction level with the distance learning system scored 4.4 out of 10. According to the evaluation regarding whether the level of satisfaction of participants varied across the type of university, the satisfaction level of students in public universities is generally low (score of 4.3 out of 10) compared to satisfaction in foundation universities (4.9 out of 10).
